When you reach Jacobs Estate, there would be one huge task to finish and only once you do, the game allows you to continue moving further to get the Vault Key clue. Otherwise, all the doors that lead there will be locked, and you will be left spinning around trying to go through without any luck with that task.
To be able to kill Anointed Goliath, make sure you have one powerful weapon with over 1000 damage. There will be all sorts of distraction, you will be attacked many times before you get the chance to engage in fight with that Goliath, so make sure that you spare the best weapon for last, and don't wait a moment, start shooting, then use your skills, and run to hide and recover your shield. Then repeat, and don't back during the second wave, change position from higher to lower to avoid damage, go up and down stairs, and once you see his health dropped to one third, keep shooting even if you lost your shield.
